49 IMMORTALISED CELL LINES FOR VIRUS DIAGNOSIS. J. Clarke*, C. MacDonald+, U. B. Griffiths*. * PHLS CAMR, Porton Down, Salisbury, UK. + University of Paisley, Paisley, UK. Abstract. No single established cell line has been found capable of substituting for primary baboon kidney (PBK) cells in human virus diagnosis. A panel of cell lines could be assembled which detected the most important viruses routinely encountered, but was less sensitive than PBK cells (1). Oncogene transfection has now been successfully used to immortalise PBK cells, and some of the resulting cell lines have retained virus susceptibility.

Introduction The productivity of the cell line selected for the manufacture of a biopharmaceutica l has a major influence on the economics of the production ofthat molecule. The dominant paramete r for suspensio n batch cultures is product concentration . Following transfection, cloning or re-cloning of recombinant mammalian cell lines it is common practice to select potential cell lines for such manufacturing processe s according to the following scheme : 1, Assay the concentratio n of product produced by over 100 cell lines whilst they are growing in the initial 96-well tissue culture plates and select the highest producers .
